اثر همزمان محدودیت غذایی و مهار گیرنده D2 دوپامینی بر حافظه فضایی موش‌های صحرایی نر

زمینه و هدف: محدودیت غذایی می‌تواند اثرات مفید یا مخرب بر عملکردهای مغز از جمله یادگیری و حافظه داشته باشد. همچنین گیرنده‌‌‌های دوپامینی در فرآیند یادگیری و بازخوانی اطلاعات دخالت دارند. این مطالعه به منظور تعیین اثر همزمان محدودیت غذایی و مهار گیرنده D2 دوپامینی بر حافظه فضایی موش‌های صحرایی انجام شد.

روش بررسی: در این مطالعه تجربی 60 سر موش ‌صحرایی نر نژاد ویستار در 6 گروه 10 تایی شامل کنترل، محدودیت غذایی 25درصد، 50درصد، 75درصد، سولپیراید (آنتاگونیست گیرنده D2، mg/kg/day ip4)، محدودیت غذایی 75 درصد دریافت کننده سولپیراید قرار گرفتند و به‌مدت 21 روز تیمار شدند. برای سنجش حافظه از ماز شعاعی هشت‌بازویی استفاده شد. سپس میزان کاتالاز و مالون‌دی‌آلدهید هیپوکامپ‌ اندازه‌گیری گردید.

یافته‌ها: محدودیت غذایی 25درصدی منجر به کاهش 11.8 درصد زمان سپری شده برای یافتن غذا در مقایسه با گروه کنترل گردید (P<0.05). محدودیت غذایی 75درصدی و یا تزریق سولپیراید این زمان را به ترتیب به میزان 24.4 درصد و 18.3 درصد به‌طور معنی‌دار افزایش داد (P<0.05). گروه 75درصد محدودیت غذایی دریافت کننده سولپیراید نسبت به تمامی گروه‌‌‌ها بیشترین افزایش معنی‌دار زمان یافتن غذا را نشان دادند (P<0.05). فعالیت کاتالاز فقط در گروه‌های محدودیت غذایی 75درصد به‌میزان 17.6 درصد و 22.2 درصد کاهش معنی‌دار نشان داد (P<0.05). میزان تولید مالون‌دی‌آلدهید در گروه‌های محدودیت غذایی 75درصد به‌میزان 50.2 درصد و 59.3 درصد و دریافت کننده سولپیراید به‌میزان 31.2 درصد به طور معنی‌دار در مقایسه با گروه کنترل افزایش یافت (P<0.05).

نتیجه‌گیری: همزمانی اعمال محدودیت غذایی و مهار گیرنده‌‌‌های D2 دوپامین، منجر به افزایش سطوح پرواکسیدانی هیپوکامپ و تشدید اختلال حافظه موش‌های صحرایی می‌گردد.
