Abstract

Abstract Background and objectives: Tuberculin is the proteins existed in tuberculosis culture medium which precipitated by trichloroacetic acid (TCA) or ammonium sulfate. Tuberculin is used for diagnosis of Tuberculosis. The aim of this study is to compare the human tuberculin produced by Razi Institute and Mycobacterium tuberculosis Culture Filtrate Protein. Material and Methods: Initially By biphasic medium, Bacteria from Lowenstein–Jensen solid medium was transferred to a Dorset−Henley Liquid medium. After 6 weeks of growth, the bacteria were isolated from liquid medium containing secretory proteins by the 0, 22 micron filter and the solution containing secretory proteins was precipitated by TCA and ammonium sulfate, separately. Then, using spectrophotometer and kjeldahl protein assay, the presence of protein in solution was confirmed. At the end, the precipitated proteins are compared with the human tuberculin by Coomassie-Blue stained SDS-PAGE Results: The protein samples precipitated by TCA have more bands in the limit of higher than 20 kDa, but the protein samples by ammonium sulfate have more bands in the limit of less than 20 kDa. Human tuberculin proteins are like smear and their weight is less than 16 kDa. Conclusion: It seems that ammonium sulfate is more suitable for low molecular weight proteins than TCA for precipitation. Key words: Mycobacterium tuberculosis, SDS-PAGE, tuberculin

Abstract

Abstract Background and Objective: One of the diabetes complications is the tissue damage caused by the imbalance of oxidants and antioxidants (oxidative stress). The aim of the present study was to evaluate the activity of two antioxidant enzymes -superoxide dismutase and catalase- in the serum of streptozotocin-induced diabetic rats. Material and Methods: This investigation was conducted on adult male rats assigned to diabetic and control groups. Diabetes was induced by a single intraperitoneal injection of streptozotocin. Seven weeks after diabetes induction, glucose concentration, superoxide dismutase and catalase activities of the serum were assessed. Results: Glucose concentration of streptozotocin-injected animals was significantly higher than that of control group (P<0.001). The level of Serum superoxide dismutase and catalase activities in diabetes group were significantly higher than those in control group (P<0.01). There was a positive significant correlation between glucose concentration and superoxide dismutase and catalase activities (P<0.001). Conclusion: The high activity of antioxidant enzymes in diabetic rats is probably due to compensation responses to oxidative stress produced by high concentration of free radicals. Abstract

Abstract

         Background and Objectives: Concentration low-density lipoprotein (LDL) is one of the strongest indicators of atherosclerosis and predicts the diagnosis of cardiovascular diseases. LDL measurement accuracy is very important. LDL can be measured directly, such as enzymatic and nephelometry methods or can be calculated using Friedewald's formula. Despite the development of enzymatic methods and LDL nephelometry still in most laboratories is calculated using Friedewald's formula. The aim of this study was an investigation of correlation coefficient between two methods of measuring LDL- cholesterol levels.

         Methods: This descriptive cross-sectional study, performed on the 1141 patients. Cholesterol, triglycerides, HDL, LDL all patients assayed by enzymatic method. For patients with triglyceride levels of less than 400 mg/dl had LDL levels were calculated by Friedewald's formula. Normal levels of LDL/HDL ratio of less than 3.5 were considered.

          Results: Of the 1141 patients participating in this study, 38.3 % men and 61.7 % women. The mean patient age was 46.3 ± 16.1 years. Mean serum cholesterol, triglycerides and HDL were 177.9 ± 41.1, 132.9 ± 73.2 and 45.8 ± 13.2 mg/dl, respectively. Average direct and calculated LDL concentration was 82.1 ± 23.1 and 105.5 ± 35.8, respectively. The direct measurement of LDL, LDL/HDL levels in 97.1% of cases was normal, while 85.1 % of the calculation of LDL were normal. Pearson correlation coefficients were obtained by two methods: 0.869 (p <0.001).

         Conclusion: Despite the favorable correlation between two methods of measurements of LDL, the results of a calculation method is more than direct method. This can have a negative impact on the judgment of the treating physician.

Abstract

ABSTRACT

       Background and Objective: Measurement of amino acids is an important tool for metabolic studies and evaluation of patients’ clinical condition. The aim of this study was to analyze the plasma amino acids using reverse phase high performance liquid chromatography techniques (RP-HPLC) with pre-column derivatization by o-phthalaldehyde (OPA) in combination with 3- mercaptopropionic acid (3-MPA).

       Methods: Overall, 107 neonates and babies suspected of having metabolic disorder were enrolled in this study. The level of amino acids in plasma samples was analyzed within 65 minutes by HPLC with pre-column derivatization by OPA/3-MPA. This was a gradient RP-HPLC method that was performed using two solvents with a ratio of methanol and sodium acetate. L-norvaline internal standard was used as the reference peak for amino acids. Standard mixture of amino acids was used to determine the concentrations of amino acids.

        Results: According to the values of coefficient of variation obtained for each amino acid, the results indicated a good chromatographic separation of amino acids by this method. The use of OPA/3-MPA derivative reagent increased the efficiency and resolution of amino acids chromatographic separation.

      Conclusion: Due to simple preparation and accurate assessment, determination of plasma amino acids using OPA/3-MPA derivatives and RP-HPLC is a suitable method in many clinical samples.

Abstract

ABSTRACT
        Background and Objectives: The unfavorable lipid profile in obese individuals is associated with high incidence of various diseases including cardiovascular disease, hypertension, etc. Dieting for weight loss and physical activity are among the most important factors affecting the serum lipid profile. The aim of this study was to compare the effect of diet with and without physical activity on body mass index and serum lipid profile of obese women.
      Methods: This clinical trial was performed in 2011 on 39 obese women referred to a weight loss and nutrition counseling center. Subjects were randomly divided into an active group (diet and exercise) and inactive group (diet without exercise). Blood sampling was done before the intervention and two months after the intervention. Data was analyzed using SPSS-16 and t-test.
       Results: Mean level of triglyceride reduced significantly in both groups. Mean level of cholesterol and low-density lipoprotein decreased significantly only in the active group. The Mean level of high-density lipoprotein in the two groups had no significant difference.
Conclusion: The results indicate that dieting for weight loss along with short-term physical activity improves serum levels of cholesterol and low-density lipoprotein, but does not affect serum HDL level.

Abstract

ABSTRACT
          Background and Objectives: The incidence rate of esophageal cancer (EC) is high in north and northeast of Iran. Taurine is a sulfur-containing amino acid with a wide range of vital biological functions. The aim of this study was to compare plasma levels of taurine between patients with EC and healthy controls.
        Methods: Plasma samples from 36 recently diagnosed cases of EC and 36 healthy adults were analyzed by high-pressure liquid chromatography. Data analysis was done using SPSS (version 16). The t-test was used to evaluate significant differences in the level of taurine between the two groups. P-values less than 0.05 were considered statistically significant.
         Results: Patients with EC had significantly higher levels of plasma taurine compared with the controls (P<0.05).
         Conclusion: Patients with EC have significantly higher levels of plasma taurine compared with healthy individuals. This finding suggests that the measurement of plasma taurine in patients with EC could be useful for the early diagnosis of the disease.

Abstract

Background and objective: Human breast milk is the primary food for newborns that possess all essential nutrients for their growth and health. However, breast milk can be contaminated with various toxins including aflatoxin M1 (AFM1), a hydroxylated metabolite of aflatoxin B1 formed in the liver and excreted into the breast milk. This toxin can have immunosuppressive, mutagenic, teratogenic and carcinogenic effects. The present study aimed to investigate the level of AFM1 in human breast milk samples from Jiroft (Kerman Province), Iran.
     Methods: A total of 84 human breast milk samples were collected from lactating mothers who were referred to number one clinic in Jiroft from April 2016 to January 2017. The level of AFM1 was measured using commercial enzyme-linked immunosorbent assay kits.
      Results: Aflatoxin M1 was detected in all (100%) human breast milk samples within the interval values of 3.2 to 8.8 ng/L (mean, 4.1±0.7 ng/L). The level of AFM1 in all samples was lower than the maximum tolerable limit (25 ng/L) suggested by the EU and the Codex Alimentarius Commission.
      Conclusion: Although AFM1 is present in all human breast milk samples from the city of Jiroft, the level of this toxin is within the tolerable limit. Therefore, it seems that infants are not at risk.

Abstract

Background: Klinefelter syndrome (KS) also known as 47, XXY is one of the most prevalent chromosomal abnormalities among men. Infertility is one of the most primary features of this condition. However, there are some other associated features such as thin and tall appearance, absent, delayed or incomplete puberty, small and firm testicles, small penis and gynecomastia.
Case description: We herein report a patient with mosaic KS whose karyotype consisted of 47, XXY/46, XY. The case’s wife had two miscarriages, followed by a healthy girl with a normal karyotype who was born taller than the average at the age of two.
Conclusion: Mosaic KS dramatically increases the chance of having healthy offspring with normal genetic patterns without performing artificial insemination methods compared to those with complete KS.

Abstract

The prevalence of hypothyroidism is much higher among women than in men. Hypothyroidism is also one of the most prevalent thyroid disorders among women of reproductive age. The disease exerts its effect on female sex hormones by manipulating the production of luteinizing hormone and follicle-stimulating hormone that are crucial for the production of estrogen by the ovaries. Various studies demonstrated the adverse effect of overt hypothyroidism on ovulation, menstrual cycle, and fertility. This review surveys the adverse effects of hypothyroidism on fertility and pregnancy.
