Abstract

Background & Objective: Tetra Carbon Cholride has been known as reference hepatotoxin because it can cause necrosis, fatty change, cirrhosis and cancer liver. Silymarin has hepatoprotective and anti hepatoxin effect. This study was done to determine the protective effect of Silymarin in acute hepatotoxicity of CCl4 in rats.

Materials & Methods: In this experimental study, we chose 25ml/kg dose of CCl4 (in mineral oil solvent) as an optimum dose. The hepatotoxic effects of intraperiotoneal injection of CCl4 for obtaining parameters of toxicity and therapeutic effects have been examined. According to enzymatic results (increase in ALT and AST) and histopathologic changes (grading the changes in liver including cytoplasmic granularity, cloudy swelling, necrosis and fatty change), the interval between prescribing silymarin and sampling  was determined. Silymarin as a suspension in propylene glycol CMC 2% (3/2 ratio) has been prescribed in 50, 200 and 800mg/kg doses and serum and liver samples were obtained. Negative control group received silymarin vehicle in CCl4 solvent, drug control received 800 mg/kg of silymarin in CCl4 solvent and positive control received silymarin vehicle after injecting CCl4.

Results: The results showed that prescribing 50mg/kg silymarin one hour after injecting CCl4, in addition to inhibiting transaminase activity, prevents progress of liver injury up to 50% of positive control group. Cellular repair and regeneration are also enhanced, So the grade 3necrosis in positive control group is decreased to grade 0.5 in silymarin gourp in 48 hours prescribing silymarin (50mg/kg).

Conclusion: This study showed that up to six hours after injecting CCl4 significantly prevents hepatotoxicity, and cause acceleration in repair of liver injuries.

Abstract

Background and Objective: The diagnosis of oral lesions can be more difficult than skin lesions, it either may be missed, or have no difference in color with near mucosa. The correct diagnosis is important, because treatment and prognosis either of them is different. The purpose of this study was to survey the pathological oral lesions in Ahwaz the capital city of Khozestan rpovince in South-West of Iran. Materials and Methods: This retrospective descriptive cross sectional study was done on 588 pathologic specimen, types of oral lesions, on patients referred to department of pathology, Imam Khomeini hospital in Ahwaz, since 1994 to end of 2004. Results: The mucosal oral lesions were more common in women than men (305 cases, 54%), the most lesions were benign (461 cases), the numerous lesions were nonspecific, most of the patients were in range of 20-30 years. The most common malignant tumor was squamous cell carcinoma and the rarest malignant tumor was malignant melanoma that all of them were on lower lip. The most common site for lesions was lower lip (155 cases, %27.6). The malignant cases were more common in men than women. Conclusion: This study showed the benign mucosal oral lesions were more common in female, than males. The lower lip was the most common site which is similar to other reports. Squamous cell carcinoma was the commonest malignant tumor which has a correlation with other studies.

Abstract

Background and Objective: Human erythropoietin (EPO) is a glycosylated hormone with molecular weight of about 40 KDa which is synthesized in kidneys and plays an important role in proliferation and differentiation of erythrocytes.This study was done to assess and analyze the expression of recombinant EPO in Leishmania tarentolae host. Method: In this descriptive study, the EPO gene was codoned , optimized with bioinformatics database prior to be synthesized. It was cleaved by KpnIandXbaI enzymes and cloned into pLEXSY expression vector. The constructed expression cassette was transfected into Leishmania tarentolae through electroporaton method. Identification and confirmation of transfected colonies was performed using PCR expression diagnostic primers and EPO specific primers. Induction of the cloned gene was done with tetracycline. The expression in induced strains was analyzed by SDS-PAGE and western blotting techniques. The amount of recombinant protein was quantified by ELISA method. Confirmation of cloning and EPO expression cassette was carried out through genetic engineering procedures. Results: Expression analysis of transfected parasitic strain with SDS-PAGE and western blotting confirmed gene integration into chromosomal of host as well as expression. The optimal conditions for expression were found to be 10μg of tetracycline and 72h induction time. Molecular weight of expressed protein estimated to be 40 KDa and expression level was determined to be 12.4 mg/l which was equal to 1% of total protein mass. Conclusion: EPO expression cassette for cloning and expression in Leishmania tarentolae was designed and protein of interest was successfully induced and identified Leishmania tarentolae can be used as a suitable host for production of recombinant EPO and this technology has a potential for localization.

Abstract

Background and Objective: Biofilms caused by pathogenic microorganisms that plays an important role against human health. Due to their resistance to detergents and antimicrobial agent, treatment response of affected patients with these bacteria is difficult. This study was done to evaluate the effect of methanol extract of Nasturtium officinale plant on growth and biofilm formation of Pseudomonas aeruginosa.
Methods: In this descriptive - laboratory study, the extraction was done by Maceration in 80% methanol and by rotary evaporator. The minimum inhibitory concentration (MIC) of Nasturtium officinale extracts were determined by broth microdilution method. Biofilm formation was investigated using the microtiter plate and stained with crystal violet.
Results: The minimum inhibitory concentration of Nasturtium officinale against Pseudomonas aeruginosa was 0.625 mg/ml and the Minimum bactericidal concentration of this extract was 1.25 mg/ml. PAO1 strain and 5 clinical strains were able to biofilm formation. Inhibition of biofilm formation by extract of Nasturtium officinale plant was dependent to concentration. The highest percentage of inhibition of biofilm formation was in the concentration of 7.5 mg/ml and the lowest percentage of inhibition of biofilm formation was in the concentration of 0.11 mg/ml. The mean of Pseudomonas aeruginosa biofilm inhibition by Nasturtium officinale extracts was 72.69±22.27 %. In the concentrations of 7.5, 0.93, 0.46, 0.23 and 0.11 mg/ml, there was a significant difference between clinical strains and PAO1 strain (P<0.05).
Conclusion: Methanolic extracts of Nasturtium officinale plant has anti-bacterial and anti-biofilm effect against Pseudomonas aeruginosa.

Abstract

Background and Objective: Increased ferritin levels have been implicated in the pathogenesis of diabetes. This study was done to determine the effect of continuous and interval aerobic exercise training on serum ferritin and iron level of induced diabetic rats.
Methods: In this experimental study, 32 wistar rats weighing 205±54 g were randomly allocated into four groups including healthy control, diabetic control, diabetic interval training and diabetic continuous training. The diabetic training groups were received 10 weeks of training following one week after the induction of diabetes by streptozotocin and nicotinamide. Fasting blood sugar ferritin and iron level was measured in each animal.
Results: Induction of diabetes significantly increased serum ferritin and iron levels in diabetic control group compared to healthy control group (P<0.05). Serum ferritin and iron levels and fasting blood sugar significantly reduced in interval aerobic exercise and continuous aerobic training groups compared to diabetic control group (P<0.05). There was not any significant difference in the dependent variables between interval aerobic exercise and continuous aerobic training.
Conclusion: In conclusion, it seems the training of two methods of aerobic interval and continuous training in induced diabetic rats with reduction of serum ferritin and iron levels, as well as improving fasting blood glucose and serum insulin seems to reduce insulin resistance index and improve glycemic status in induced diabetic rats.
