Abstract

Streptococcal pharyngitis and rheumatic fever/rheumatic heart disease are among the major public health problems in children. In this study we assay the frequency of group A Beta hemolytic Streptococcus in nasopharynex of 1588 healthy 6-12 years children in Gorgan in north of Iran. The sample obtained during February to March 1998, using sterile soap. The prevalence of this bacteria were 11% (175 strains) the rate of colonization in girls and boys were 10.8 and 11.2% respectively. Our study shows that the carrier of this bacteria in Town’s school children are more than the villages.

Abstract

Golden Staphylococci is one of the most common factors in skin and systemic infection. About 30% of people are carrying this bacterium in their skin and nose. Increased drug resistance in this microorganism and consequent infection, has gained the attention of scientific organization. Therefore we decided to determine the prevalence of nose carrier of this bacterium in healthy children in Gorgan schools. During the winter 1998 to March 1999 the sample has been taken from 1193 student’s nose with sterile cotton swab. These results showed that in 194 samples (16.3%) Staphylococci Aureus has been isolated, from these samples 114 cases belong to girls (16.1%) and 80 cases were boys (16.4%) that there is not a meaningful variation between them. The number of bacteria isolated from the villages children was 109 cases (17.6%) and in town’s children were 85 cases (14.8%), also this difference didn’t show significant difference. The prevalence of carrier among the 6-12 years old was 12-25.3%, which has got a significant meaning (P<0.001). Antibiotic resistance in the isolated S.aureus were tested and result showed that 34.8% of samples are resistant to Methicilin and 1.7% were resistant to Vancomycin, only 4% were sensitive to Penicillin. Therefore with regard to the results of this investigation, further researches are recommended in relation between strain’s of S.aureus that isolated from carrier’s and strain’s that isolated from patients and to determine their drug resistance in this region.

Abstract

Background and Objective: Lectins are carbohydrate-binding proteins of plants or animal origin, which react with terminal sugars of glycoconjugates of cells and tissues. Glycoconjugates play essential role in several biological phenomena such as Cell recognition, proliferation, migration and differentiation. Notochord, a mesodermal derivative, induces nearby tissues, including Neural tube, sometimes and developing gut. Materials and Methods: The purpose of this study was to investigate distribution of glycoconjugates in component of notochord and axial vessels during mouse embryonic development. Results: Formalin fixed sections embedded in paraffin from 9 to 14 embryonic days of Balb/c mice were processed for histochemical studies by using four different horseradish peroxidase (HRP) labeled lectins including: Glycin Max (SBA), Vicia Villosa (VVA) and Maclura Pomifera (MPA) specific for GalNac terminal sugar and Lotus Tetragonolobus (LTA) that bind to ?-L-fucose terminal sugar. Our results revealed that approximately similar reactions were observed in notochord and axial vessels endothelium by using GalNac sensitive lectins during vessels morphogenesis. Fucose sensitive lectin reacted only with the notochord. The time and significant role(s) in tissue interactions and subsequent formation of adjacent tissues such as: Axial vessels development during critical morphogenic period. Conclusion: Furthermore, our findings indicate that fucosylated glycoconjugates may have no role in axial vessel formation and/or their interactions with the notochord during early morphogenic period.

Abstract

Background & Objective: Neural tube defect is one of the most important malformations of the newborns. The aim of this study was to investigate the rates of NTD, anencephaly, spina-bifida cystica, encephalocele and the relation of these abnormalities with sex, ethnicity, mother’s age and consanguineous marriage in north of Iran (Gorgan). Materials & Methods: A cross-sectional study was done on 26280 birth in Dezyani Hospital in Gorgan during 1998-2001. Results: This study has showed that, the NTD rate was 3.08, 2.78 and 3.38 per 1000 in total, males and females, respectively, female to male ratio was 1.31. The rate of spina-bifida, anecephaly and encephalocele were 1.48, 1.48, 0.11 per 1000, respectively. In regard to different races, the rate was 6.78/1000, 4.76/1000 and 2.41/1000 among Turkmans, Systani and native Fars, respectively (P<0.05). The rate of NTD in the mothers under 20 years was 2.29/1000, in 20-34 years was 3.37/1000 and over 35 years was 2.54/1000. Interstingly, 29.6% of parents with affected newborns had consanguineous marriage. Conclusion: We concluded that there is a higher rate of NTD in this, area, that it might be due to race-ethnicity variations.

Abstract

Background & Objective: The pattern of cell types in vertebrate embryos depend on the function of organizing centers, specialized cell groups that direct the fate of nearby cells through the secretion of inductive factors. Our previous studies showed that during early neural tube formation, the notochord is essential for the induction of ectoderm and for the subsequent differentiations of the neuroepithelium. It is well known that glycoconjugates are developmentally regulated expression on the surfaces of early embryonic cells and could therefore be involved in many critical morphogenetic and histogenetic events during embryonic development. Materials & Methods: In the present study, histochemical studies were carried out to detect the presence and distribution of terminal sugars during development of precursors of motor neurons within the developing spinal cord in balb/c mice. Embryos from day 9 to 14 of gestation were fixed and processed for lectin histochemical studies by using horseradish peroxidase labeled WFA with binding specificity for terminal N-acetylgalactosamine. Results: The first reaction was occurred weakly on the cells surface and extra cellular matrix just around the peripheral portion of floor plate on day 13 of gestation. It seems that these cells are developing premotor neurons, which will form the future motor neurons of the spinal cord. The reaction increased significantly and extended to the deep part of spinal floor plate by day 14. Conclusion: These data indicate that glycoconjugates containing N-acetylgalactosamine may play important roles in differentiation of the floor plate motor neuron and perhaps glia cells in final development of the ventral part of the spinal cord.

Abstract

Background&Objective: Carbohydrate chains link to glycoproteins and glycolipids of the cell surface particulary their terminal sugars have key roles in cellular activities of almost all biological systems. Distribution of these glycoconjugates in adult kidney has not received fair attention yet. The purpose of this study was to identify in situ location of galactose – N – acetyle galactoseamine moleucles on the surface of the adult rat kidney. Materials&Methods: After the resection of rat kidney and rotin fixation with regular lectin histochemical technique, MPA and PNA conjugated to horseradish peroxidase, lectins specific to indentify these terminal sugars in rat kidney. Results: Intense reaction of PNA to collective tubules, less intense and reaction with luminal surface of proximal tubules and MPA identified distinct cells among distal tubules as well as glomerules were observed. Conclusion: It seems that this specific sugars related to the cellulars function of this region of adult kidney and probably genetically is regulated during embryonic development.

Abstract

Background & Objective: Preterm infants have less iron storage compared with the term one. Due to rapid growth they need more iron during infancy. This study was designed to evaluate the effect of early iron supplementation on hematologic indices and incidence of iron deficiency anemia in preterm infants. Materials & Methods: This experimental study was done on 20-days-old preterm breast-feed infants referred to the Gorgan-North of Iran primary health care services for vaccination, without any underline disease or growth retardation during 2005. They were divided to two groups (n=15 in each) and a questionnaire was completed for each case. Iron drop (2mg/kg) was given in the interventional group. Iron deficiency anemia was assesed in all sample T-student test and chi-square were used to analyze the independent variables and comparing the hematologic indices, after entering in SPSS-13 software and testing the normal distribution with komologrof-smearnoff test. Relative risk index (RR) was used to compare the two groups. Results: The relative risk of iron deficiency anemia in the interventional group was 0.4 folds [RR=0.4,CI %95 for RR=(0.091,1.749)]. No significant difference was seen between the two groups. Conclusion: The finding of this study showed that iron drop supplementation for preterm infant prevent the iron defeciency anemia, although this observation was not significant.

Abstract

Background & Objective: Health evaluation and growth monitoring of the infant is a visual diagram of growth and primary health services are based on it. In the best option, these indices must be provided separately for each community. This study was done to evaluate growth indices of newborns in Gorgan- Northern Iran. Materials & Methods: In this observational descriptive study which was carried out during (Oct-Nov 2003), all healthy infants without any major abnormality born within 25-42 weeks of gestational age were evaluated in Dezyani Hospital, Gorgan growth indices (head circumference, length and height) were measured in 1011 infants. Gestational age was defined by Balard scoring and delivery type was recorded, too. After entering data into SPSS-10 software, data were analyzed using chi-square, t-test and correlation bivariate. Results: Mean gestational age (±SD) was 39.86±1.72 weeks and most of them were born by vaginal delivery. Amongst them, 3.2% were premature (less than 37 weeks), 4.4% were low birth weight and 0.69% were very low birth weight. Correlation between growth indices and gestational age was significantly positive in all cases (p<0.05). Conclusion: This study showed that growth indices are lower than other parts of Iran. Further investigations needed, especially longitudinally and cross-sectional, to determine the growth indices in this region and in other parts of country.

Abstract

Background and Objective: Change in the cell surface and extracellular matrix glycoconjugates has been reported in many cancers. Moreover, diagnostic and prognostic importance of these substances and also their roles in therapeutic modalities for cancerous patients has been emphasized. This study was designed to explore the histochemical study of cellular mucopolysaccharides in esophageal and gastric carcinoma and its relation to tumor differentiation. Materials and Methods: In this laboratory study tissue samples of 40 patients with esophageal squamous cell carcinoma and 40 patients with stomach adenocarcinoma in different grades of tumor were selected from pathology department of Emam Reza hospital in Mashhad, Iran. Tissue samples were stained with Alcian Blue (PH 1 and PH 2.5) for Sulfated and Carboxylated mucosubstances respectively, along with positive and negative controls. Results: Normal esophageal epithelium and carcinoma cells of different grades showed negative reactivity but normal and tumoral stromal cells depicted positive staining in both PHs. In PH 1, normal glandular and carcinoma cells of the stomach were negative but in PH 2 glandular cells were positive though carcinoma cells showed weakly staining. Normal and tumoral gastric stromal cells showed positive staining in PH 1 and PH 2.5. Conclusion: It is highly probable that in the process of cancerization of normal esophageal squamous cells, functional changes, from the perspective of producing Carboxylated and Sulfated mucosubstances, do not occur, whereas some changes in glandular cells of stomach which result in diminishing the production of Carboxylated mucosubstances during cancerization process are observable.

Abstract

Background and Objective: The neurohypophysis originates from the floor of diencephalon. Its development controles by several cellular interactions that mediated by some molecules such as cell surface and extra cellular matrix Glycoconjugates terminal sugars. In this study we used lectin histochemichal technique to evaluate distribution of the Glycoconjugates and their changes during development of neurohypophysis. Materials and Methods: This experimental study carried on 40 female and 20 male adult Rats. After mating and appointment day zero of pregnancy, pregnant Rats were sacrificed from days 10-20 of gestation and their embryos were collected for histochemical study. The serial section of head specimens were fixed and incubated with different HRP-lectins from Orange fungus (OFA) Vicica villosa (VVA), Glycine max (SBA), Wistaria floribunda (WFA), peanut (PNA), Griffonia simplicfolia (GSA1-B4), Lotus tetragonolobus (LTA) and Ulex Europeus (UEA-1). OFA, LTA and UEA-1 lectins are specific for terminal sugars α-L–Fucose and WFA, SBA, VVA and PNA are specific for D-GalNAc, α, ß-D-GalNAc and GalNAc, D-Gal-(ß-1-3)- D-GalNAc of complex glycoconjugates respectively. Results: Our findings demonstrated that the reaction of neurohypophysis cells with OFA initiated from gestational GD10 and increased to GD15 (P<0.05) and then increased to GD17 (P<0.05). A few cells of neurohypophysis reacted with PNA from GD13 to GD16 and decreased afterward (P<0.05). Some cells of neurohypophysis reacted with SBA from GD14 to GD18 and decreased afterward (P<0.05). Reacting of many cells of neurohypophysis with WFA started on GD13 and increased to GD15 (P<0.05) and then decreased afterward (P<0.05). Neurohypophysis cells showed no reaction with the UEA-1, LTA, VVA and GSA1-B4 lectins. Conclusion: The expression of Glycoconjugates with terminal sugars α-L–Fucose, α, ß-D-GalNAc and D- Gal– (ß-1-3)- D-GalNAc have importanct role and special spatiotemporal situation in neurohypophysis development.

Abstract

Background and Objective: Continuous antigenic variation of Influenza a viruses causes a major concern to develop Influenza vaccine. Conserved antigens are suitable candidates for vaccine production due to its non-requirement to match the designed strains with circulating strains. The M2 gene is conserved among Influenza a viruses and has potential to be considered as a universal vaccine. This study was designed to evaluate the effects of aqueous Echinacea purpurea extract on immunogenicity of DNA vaccine encoding M2 gene of Influenza virus. Materials and Methods: This interventional study was carried out on female BALB/c mice with 3-4 week age (250-300 gr). Plasmid DNA encoding M2 gene (pcDNA-M2) of Influenza virus A/New Caledonia/20/99 (H1N1) was transformed into E.coli top10 f' and cultured in LB broth media. Large scale plasmid preparation was done and the concentration was measured by spectrophotometric method. Mice were divided into eight groups and immunized three times with fifteen days apart. Vaccine groups received inactivated Influenza virus or pcDNA-M2, alone or in combination with Echinacea extract. Control groups were injected pcDNA, Echinacea extract, and phosphate buffer. All animals were left to bleed before immunization and at 21 days after the last vaccination and specific anti-M2 antibodies were measured by indirect ELISA. Then the mice were intranasally challenged under an aesthesia with mouse-adapted PR8 Influenza virus and monitored for 3 weeks to evaluate the vaccine regimen efficacy in reduction of mortality rate compared to control groups. Data were analyzed using SPSS-16, One-way ANOVA and Kaplan–Meier tests. Results: The highest specific immune response was obtained in mice received inactivated virus plus extract (P<0.05). Immune responses in mice inoculated with pcDNA-M2 were significantly higher compared to all control groups mice (P<0.05). In addition the specific immune responses in group inoculated with pcDNA-M2 and aqueous extract was higher compared to the group receiving only pcDNA-M2 (P<0.001). The highest survival rate was observed in mice injected with inactivated virus or pcDNA-M2 plus extract. Conclusion: This study showed that pcDNA-M2 induced specific immunity and protected mice against lethal challenge with PR8 Influenza virus. Furthermore, application of Echinacea extract with M2 gene vaccine increased vaccine efficacy.

Abstract

Background and Objective: Different organizers are involved in spinal cord development and differentiation by sending various messages. Specific glycoconjugates secreted from the cells of lateral wall of spinal cord can also act as neurogenesis and neural differentiation messengers. This study was carried out to determine the distribution of sugar compounds in the lateral walls of spinal cord during mice morphogenesis using lectin histochemistry method. Methods: In this experimental study, sections of BALB/c mice from 10-16 embryonic days were fixed in formalin and then histological sections were prepared. Tissue samples for reaction to the glycoconjugates were incubated with DBA, OFA, GSA1B4 and MPA lectins. Alcian blue with pH equal 2.5 was used for background staining. Results: DBA lectin did not react with the lateral wall of the spinal cord. MPA lectin showed severe reaction but consistent, especially in nerve fibers of the lateral wall of spinal cord. GSA1B4 lectin showed weak reaction in the cells and nerve fibers of the spinal cord, but severe reaction was clearly observed in blood vessels. OFA lectin showed severe reaction with α-L-Fucose terminal sugar in the lateral walls of the spinal cord in early stages of morphogenesis. Conclusion: The most reaction in the lateral walls of the spinal cord was related to OFA, which reflects the importance of fucose terminal sugar by connecting (1→6) to the penultimate sugar N-acetyl-D-glocosamin (Glc-Nac) in the development of spinal cord. Due to severe reaction of GSA1B4 to blood vessels of spinal cord, use of this lectin for vascular studies, is recommended.
