Abstract

Abstract Background and objectives: Microbial, physical and chemical properties of drinking water, considering type and quantity of the drinking water, are the factors for determining the acceptability of water. The effect of each of the above components on increasing the quality of water or threatening the health of consumers is also considerable. We aimed to investigate the trace amounts of microbial, physical and chemical parameters of drinking water in Gorgan, comparing to national and international standard. Material and Methods: In this cross sectional study, 50 samples were taken, per month, from four pilot stations in urban water network of Gorgan during one year (2009- 2010). We used instant sampling and performed Enumeration of Coliforms in water culture with most probable number (MPN).Samples taken from the stations during the month were investigated in two general categories of device test and heading-meter. All experiments were performed using standard reference method. Results: of 598 water samples 551 (92.2%) have not Coliforms, 47 (7.8%) contain Coliforms and 16 (2.7%) of them have thermo-tolerant Coliforms. Mean water hardness is 413mg/L, base on carbonate calcium. The amount of fluoride about 0.35 mg and the residual chlorine about 0.6 mg is less than national standard amount. Conclusion: The results show that Gorgan water network is, in view of microbial contamination, safe during almost all days. All the tested samples in terms of physical and chemical parameters are in the range of nation and international standards. Hardness of drinking water of Gorgan is one of the highest. Abstract

Background and Objective: West Nile virus (WNV) is a member of the genus Flavivirus that can cause viral infections in human. This study aimed at detecting IgG antibodies against WNV in patients of two cities of Neka and Shiraz.

Material and Methods: the participants were 46 possible WNV case from Neka (13 women and 10 men) and Shiraz (10 women and 13 men).  IgG assay was carried out using the Elisa method.

Results: Immune Status Ratio (ISR) in Neka was negative for WNV IgG, but 12 from Shiraz, including 2 women and 10 men, were positive for WNV IgG that was changed from 3.12 to 38.6. Considering ISR, there was significant variation between Neka and Shiraz (p<0.05). In addition, results showed that there was significant variation in WNV infection rate between men (39.19%) and women (13.04%) from Neka and Shiraz cities (p<0.05).

Conclusion: Because Shiraz has hot and semi-dry climate, whereas Neka temperate climate, the results were affected by climate variation.  Given the outdoor job of men compared to women, they are exposed to the bite of mosquito vectors that transmit WNV.

ABSTRACT
Background and Objectives: The incidence rate of esophageal cancer (EC) is high in north and northeast of Iran. Taurine is a sulfur-containing amino acid with a wide range of vital biological functions. The aim of this study was to compare plasma levels of taurine between patients with EC and healthy controls.
Methods: Plasma samples from 36 recently diagnosed cases of EC and 36 healthy adults were analyzed by high-pressure liquid chromatography. Data analysis was done using SPSS (version 16). The t-test was used to evaluate significant differences in the level of taurine between the two groups. P-values less than 0.05 were considered statistically significant.
Results: Patients with EC had significantly higher levels of plasma taurine compared with the controls (P<0.05).
Conclusion: Patients with EC have significantly higher levels of plasma taurine compared with healthy individuals. This finding suggests that the measurement of plasma taurine in patients with EC could be useful for the early diagnosis of the disease.

ABSTRACT
Background and Objectives: Blood transfusion may induce some adverse effects on receivers. Some methods such as antibody screening and cross matching have been suggested to reduce the risk of transfusion complications. However, these methods require commercial antibody screening kits that may also need special equipment. The aim of this study was to introduce a new method for antibody screening that does not require a commercial kit, and could be used in any transfusion laboratory.
Methods: We examined 350 samples that contained alloantibody and 350 control samples without the antibody. A solution containing two O+ and one O- samples were used instead of screening cells.
Results: Sensitivity and specificity of the method were 73.32% and 45.15%, respectively. Positive predictive value and negative predictive value were 58.33% and 63.88%, respectively.
Conclusion: Our new method can be used in basic hematology laboratories with some modifications.
      Keywords: Antibodies, Antigens, Coombs test.

ABSTRACT
Background and Objectives: Diabetes is one of the most common diseases and a major risk factor for cardiovascular disease. Studies have shown that regular exercise can affect apoptosis in cardiomyocytes. The purpose of this study was to investigate the effect of high-intensity interval training on the expression of Bcl-2 and Bax as important apoptosis factors in diabetic rats.
Methods: After inducing diabetes in 20 male Wistar rats (weighing 250 ± 1.15 g), the rats were randomly divided into a control group and a training group. The training group performed high-intensity interval training five days a week for four weeks, and the control group did not perform any training. After the intervention, RNA was extracted and TCF mRNA was subjected to real time RT-PCR for measuring Bax and Bcl-2 expression in the heart tissue of diabetic rats. Data were analyzed by IBM SPSS Statistics V22 using independent t-test. P-values less than 0.05 were considered as statistically significant.
Results: The expression of Bcl-2 increased significantly and the expression of Bax decreased significantly after the four-week training intervention.
Conclusion: The high-intensity interval training can have beneficial effects on the expression of apoptotic genes in rats with type 2 diabetes.

Abstract

Background and objectives:  Oxidative stress is the most important pathophysiological cause of diseases. Studies have shown that exercise and supplementation with medicinal plants have limited side effects. The aim of the present study was to evaluate effects of aerobic exercise and ethanolic extract of purslane seed on ATP, O-6-Methylguanine DNA methyltransferase (MGMT), malondialdehyde (MDA) and prooxidant-antioxidant balance (PAB) levels in the heart tissue of rats poisoned with hydrogen peroxide.
Methods: In this experimental trial, 72 male Wistar rats were randomly divided into nine groups: (1) control + H2O2 , (2) aerobic exercise, (3) aerobic exercise and 50 mg/kg purslane seed extract, (4) aerobic exercise and 200 mg/kg purslane seed extract, (5) aerobic exercise and 400 mg/kg purslane seed extract, (6) 50 mg/kg purslane seed extract, (7) 200 mg/kg purslane seed extract, (8) 400 mg/kg purslane seed extract, and (9) healthy control. Oxidative stress was induced by intraperitoneal injection of 1 mmol/kg hydrogen peroxide three times a week for eight weeks. Aerobic exercise was performed three sessions a week for eight weeks, and the purslane seed extract was intraperitoneally injected daily at the mentioned doses.
Results: Aerobic exercise and purslane seed extract alone or combined significantly increased ATP, MGMT and significantly reduced MDA and PAB levels in cardiac tissue of rats exposed to hydrogen peroxide (P<0.05). Moreover, the effect of purslane seed extract was dose dependent.
Conclusion: It seems that aerobic exercise and purslane seed extract supplementation have synergistic cardioprotective effects under oxidative stress.

Abstract

Background and objectives: Fanconi anemia (FA) is an autosomal recessive disorder that usually manifest in forms of pancytopenia, hyperpigmentation, and skeletal complications. Mutation in the DNA repair regulatory genes is associated with the development of FA. Examination of chromosomal breakages when chromosomes are exposed to cross-linking agents is a common method of FA diagnosis. This study aimed to evaluate the prevalence and characteristics of patients with FA in Mashhad, northeast of Iran.
Methods: In this study, we evaluated 312 suspected FA patients who had been referred to the laboratory of Ghaem Hospital during 2014-2020. The mitomycin C method was used to identify FA-positive subjects.
Results: After the examinations, 84 patients (26.9%) were cytogenetically positive for FA. Of 84 patients, 48 (57.1%) were male and 36 (42.9%) were female. Thumb abnormality was the most common congenital anomaly (43.2%).
Conclusion: Based on the findings, males are more susceptible to FA, and thumb abnormality is the most common congenital anomaly associated with FA. Combination of clinical manifestations and genetic susceptibility in patients may contribute to a more accurate diagnosis.

Abstract

Background: Kawasaki disease (KD) is a febrile systemic vasculitis that affects children. Further research is required due to the disease's significance, consequences, and increasing prevalence. This study aimed to determine the frequency of coronary artery complications in patients with KD referred to Taleghani Hospital (Gorgan, Iran) from 2007 to 2017.
Methods: Between 2007 and 2017, 108 cases with a definitive diagnosis of KD were studied retrospectively. Data were obtained from medical records and the patients' biographical, clinical, laboratory, radiographic, and echocardiographic data.
Results: A total of 108 patients diagnosed with KD were involved in the study, 21 (19.44%) of whom showed coronary artery-associated complications. There was no significant difference in clinical and laboratory findings and coronary artery involvement between male and female groups (P-value < 0.05). Left coronary artery (LCA) involvement and the pattern of vascular involvement as ectasia and aneurysm were significantly higher in the echocardiography of the studied patients (P-value = 0.0001).
Conclusion: Kawasaki disease-related coronary artery complication is more common in children under the age of 3, particularly males, in Gorgan. The LCA is the most common site of coronary artery complications in KD children. The findings can improve the prognosis of KD patients and their related heart diseases.
