Abstract

Abstract Background and Objective: Cream, a rich dairy product, with a neutral PH and low preservation time is a suitable medium for microbial growth. Salmonella is one of the most important pathogens in causing food poisoning and human gastroenteritis. This study aimed at investigating the quality of traditional cream for the bacterial contamination. Material and Methods: In this cross-sectional study, 196 non-pasteurized cream samples were collected from 5 regions of Hamedan, Iran. After dilution in phosphate buffer and serial dilution preparation, Salmonella was transferred to Rappaport-Vassiliadis (RV) enrichment medium according to CDC guidelines. After 24 h incubation at 37 ° C, a loop was inoculated in MacConkey and Hektoen Enteric (HE) Agar. The suspected colony phenotype was examined and their identification confirmed by API-20 E. Results: The samples (29%) were contaminated with at least one kind of bacteria, Salmonella Spp (4.59%) and Yersinia Spp (2.55%). The other bacteria like Escherichia, Enterobacter, Klebsiella, and Citrobacter were also isolated. Nine samples were contaminated with two kinds of bacteria. Conclusion: The presence of bacteria such as Salmonella and Yersinia in unpasteurized cream indicates that more quality control needs to be applied to the traditional crème produced in the city by health control office of food products. Abstract

Abstract

       Background and Objective: Aluminum salts are among the most common useful additive compounds in preparation of human and animal vaccines. Aluminum phosphate and aluminum hydroxide are two additives that show good immunoadjuvant effects with many antigens. Aluminum-containing vaccines lead to a better and longer immune response compared to adjuvant-lacking vaccines. The Chromogenic methods used for determination of aluminum amounts in manufacturing centers are  time-consuming and requires some experienced technicians to obtain accurate results. This study aimed to design and validate a simple polarographic method to measure aluminum in recombinant hepatitis B vaccine.

       Methods: In this study, the effects of temperature, pH, potential range and potential scan rate on the polarographic method of measuring aluminum in hepatitis B vaccine was evaluated and  the optimal values for each of these factors were achieved.

       Results: In order to measure aluminum, temperature of 60 °C and pH of 4.5 were found as the optimal values. Implementation of polarographic method in the potential range of -0.25 to 0.1 volts had a better signal.

       Conclusion: Since the polarography method is more simple, accurate and faster than the chromogenic methods, it is suitable to be used for the measurement of aluminum in hepatitis B vaccine and it is recommended to be used in quality control laboratories for biological products.

Abstract

ABSTRACT

       Background and Objective: Esophageal cancer is the third most common type of cancer in Iran. Studies on water, soil, grains, waste and sediments in Golestan Province have shown that the environmental concentrations of antimony and strontium are increased in areas with high incidence rate for cancer. The carcinogenic effects of strontium have been reported, but carcinogenicity of antimony is still unclear. Since there is not enough information regarding the relationship between the concentration of strontium and antimony and prevalence of esophageal cancer in the province, this study aimed to evaluate serum levels of these elements in patients with esophageal cancer using atomic absorption spectroscopy.

       Methods: This study was performed on 30 patients with esophageal squamous cell carcinoma and 30 healthy matched controls. After sampling, concentrations of strontium and antimony were measured using atomic absorption spectroscopy.

       Results: Half of the esophageal cancer patients and control participants were male. The mean age of patients and controls was 66 ± 13 and 64 ± 8 years, respectively. The mean serum concentration of strontium in patients and controls was 24.18 ± 1.62 and 24.08 ± 1.46 μg/L, respectively. The mean serum concentration of antimony in patients and controls was 18.61 ± 1.48 and 18.98 ± 1.93 μg/L, respectively. No statistically significant difference was observed between the serum concentrations of the two elements in patients and controls.

        Conclusion: There is no difference between serum levels of strontium and antimony in cancer patients and healthy controls, which could be due to tissue accumulation.

Abstract

ABSTRACT
       Background and Objectives: Q fever is a zoonotic disease caused by an obligate gram-negative intracellular pathogen called Coxiella burnetii. The aim of this study was to determine the seroprevalence of anti-C. burnetii antibodies in bulk tank milk (BTM) samples of dairy cattle in west and northwest of Iran.
       Methods: Overall, 71 BTM samples (covering nearly 700 dairy cattle) were collected in autumn 2013. A commercial Q fever antibody ELISA Test Kit (Liebefeld-Bern, Switzerland) was used to identify the presence of antibodies against inactivated phase 1 and phase 2 C. burnetii antigens.
       Results: The results of ELISA test showed that 17 BTM samples (23.9%) were positive for the presence of anti-C. burnetii antibodies.
       Conclusion: This study is the first to evaluate presence of anti-C. burnetii antibodies in BTM samples from dairy cattle herds in west and northwest of Iran. The high prevalence of this pathogen highlights the need for pasteurization of raw milk and raising awareness in consumers of dairy products in these regions.

Abstract

ABSTRACT
         Background and Objectives: Klebsiella pneumoniae is one of the most important nosocomial pathogens. Its capsular polysaccharide is considered as the first and most important virulence factor of this bacterium. This study aimed to investigate the presence of capsular serotypes K1 and K2 in K. pneumoniae isolates to examine the virulence potency of the isolates.
         Methods: Overall, 65 capsulated K. pneumoniae isolates were collected from patients with urinary tract infections in Rasht, Iran. The isolates were examined using biochemical tests and CPS gene amplification using PCR. Mucoid phenotype of the isolates was determined by the string test. The presence of K1 and K2 genes was evaluated by PCR using specific primers for the genes.
         Results: Of 65 K. pneumoniae isolates, seven (10.77%) were positive for the presence of the K1 gene and four (6.15%) were positive for the presence of the K2 gene. In addition, six serotype K1 isolates (27.27%), four serotype K2 isolates (18.18%), and 12 non-K1/K2 serotype isolates (54.54%) had hypermucoviscosity phenotypes.
          Conclusion: Our results confirm the presence of the capsular serotypes in K. pneumoniae isolates, with a relatively high prevalence for the capsular serotype K1. This study clarifies the importance of rapid diagnosis and suitable treatment of infections caused by K. pneumoniae in prevention of complicated infections.

Abstract

Background and objective:  Huntington's disease (HD) is an autosomal dominant disorder that mainly affects adults. Although mutations in the IT15 gene have been known as the main cause of the disease, patients with HD like (HDL) syndrome have mutations in genes other than the IT15 gene.  In this study, we investigate the frequency of mutations in SCA17/TBP, JPH3 and PRNP genes in patients with HDL syndrome.
 
      Methods: The frequency of mutations in SCA17/TBP, JPH3 and PRNP genes was studied in 56 patients with HDL phenotype but without trinucleotide expansion in the IT15 gene. DNA was extracted from peripheral whole blood by the salting out method. PCR was performed using specific primers for each gene. PCR products were separated on polyacrylamide gel. Sequencing was performed on some samples to confirm the PCR results.
 
      Results: We found neither trinucleotide expansion in the JPH3 and SCA17, nor octapeptide insertion in the PRNP gene.
 
      Conclusion: Based on the results, Iranian patients with HDL syndrome do not have mutation in the TBP, JPH3 and PRNP genes. However, this result may be due to population differences, rarity of the mutations in the studied genes and the small number of study subjects. Therefore, studies with a larger study population that investigate other mutations, such as point mutations in the mentioned genes may help clarify the exact cause of HDL phenotype in Iranian patients.
