Abstract

Background&Objective: Both CD4+ type 1 helper (Th1) cells and CD8+ T cells play effective roles in protection against Mycobacterium tuberculosis infection. MPB51, a major mycobacterial secreted protein, induces humeral and cellular immune responses against mycobacterial infection. In addition, DNA vaccine encoding MPB51 can induce cellular immune responses and protective immunity upon challenge with M.tuberculosis. This study address to identify T-cell immunodominant epitopes on MPB51 in BALB/c mice. Materials&Methods: We cloned DNA encoding MPB51 molecule in pCI plasmid. After constructing MPB51 DNA-covered gold cartridge, BABL/c mice were immunized by using a gene gun system. Two weeks after the last immunization, the immune spleen cells were cultured in response to synthetic overlapping library peptides covering the mature MPB51 sequence or medium alone. Intracellular and cell culture supernatant gamma interferon (IFN-) production was analyzed by using flow cytometry and ELISA, respectively. Results: The findings of present study indicate that DNA vaccination can course strong mmune response only against the peptides contain 21-40 aminoacids. Further analysis with a computer – assisted algorithm permitted the identification of nine aminoacids of (P24-32) as immunodominant CD8+ T cell epitope. Conclusion: This study proved than the MHC class I-peptide (H2-Dd-P24-32) complex is recognized by (IFN-)–producing CD8+ T cells. We observed by using T-cell subset depletion that CD8+ T cells are the only P24-32-responded T-cells in BABL/c mice. The data obtained are useful for identifying cellular immune responses against TB and for designing a new vaccine against M.tuberculosis infection.

Abstract

Background and Objective: Helicobacter pylori (H.pylori) with positive Cytotoxin associated Gene A (CagA) have higher potential for pathogenesis. Cytotoxin associated Gene A (CagA) accelerate the pathogenecity of bacteria due to cytotoxin production stimulation. The aim of this study was to determine the prevalence of the anti-CagA antibody among H. pylori infected persons in Golestan province-North of Iran. Materials and Methods: This descriptive study was carried out on 676 H. pylori positive subjects in Golestan province, northern Iran during 2008. Anti CagA antibody were determined in H.pylori positive subjects. Data analyzed by SPSS-16 software and chi-square test. Results: Prevalence of anti CagA in Helicobacter pylori infected cases was 57.7% (390 cases: 179 males and 211 females) (95% CI: 53.9-61.4). According to age the highest and lowest cases of anti CagA antibody were seen in, 15-24 (63.4%) and under 5 years old (26.3%). The level of anti CagA antibody in Sistanian ethnicity group (67.2%) was more than other ethnic group. Anti CagA antibody in Rural area was more than urban regions. Sero prevalence of anti CagA antibody was highest in Minudasht twon (78%), located in East of province in comparison with Bandar Gaz (44%) in west of province. Conclusion: This study showed the prevalence of CagA positive Helicobacter pylori strains in this region is similar to other regions of Iran, Asia and Europe and higher than African population.

Abstract

Background and Objective: Due to an increase in spontaneous abortions finding a safe and secure method is inevitable. Some medicinal herbs have abortifacient properties. This study was done to determine the effect of abortifacient effect of mulberry white root in mice. Methods: In this experimental study, 50 mice dams were randomly allocated into five groups including control, sham and experimental group 1, 2 and 3. Animals in control group did not receive any substance. Animals in sham group were received normal saline, intraperitoneally. Animals in experimental group 1, 2 and 3 were received doses of 50, 70 and 90 mg/kg/bw of the alcoholic extract of root of mulberry white during the 7th to 12th days of pregnancies, intraperitoneally, respectively. At the 16th day of the pregnancy the uterine tubes of mice were removed and the absorbed fetuses were recorded. Results: The mean of absorb fetuses was 7.4, 7.6, 1.8 and 3.1 in the experimental group 1, 2, 3 and sham, respectively. There was a significant relation between the mean number of aborted fetuses and concentration of strawberry root (P<0.05). The apparent anomaly in fetus was not recorded. Conclusion: Root of mulberry white has abortifacient effect with dose-dependent manner.

Abstract

Background and Objective: Hospitalization of patients in the intensive care units frequently results in anxiety of the family members. This study was performed to investigate the effect of a family-centered clinical intervention on the anxiety of family members of the patients hospitalized in intensive care units.

Methods: In this clinical trail study, eighty family members of the patients hospitalized in the intensive care units were randomly divided into interventional and control groups, respectively. In the interventional group, the family members were allowed to be present at the bedside from the second till the sixth day.The participant's level of anxiety was assessed in the second and the sixth day of patient's hospitalization using hospital anxiety and depression scale. Oral information about the equipment and the regulations and, a daily information card consisting of treatment and care activities, vital signs and patient's level of consciousness was delivered to the participants and their questions were answered.

Results: In the the sixth day, there was a significant difference between the mean anxiety scores in the intervention (11±3.22) and control group (7.05±3.44) (P<0.05). Also, the frequncey of anxiety in the interventional group (56%) was significantly higher than the control group with 15% (P<0.05). The odds ratio for anxiety in sons/daughters was lower than other family members in the sixth day (P<0.05).

Conclusion: Providing information, presence at the bedside and participation in the care of their patient increase the anxiety of family members in the first week of patient hospitalization in the intensive care unit.

Abstract

Background and Objective: The most important factor in the integrity of saphenous vein is the health degree of endothelium which guaranties the dilation of them after cronary bypass sergery. Kind of preservative soulution has a key role in endothelial protection. This study was done to evaluate Protective effect of perserved solutions of krebs contains verapamil, adrenaline and propranololol in comparision with Krebs and Heparinated blood on desqumation of the endothelial cell in Saphenous vein of Guinea pig.
Methods: This experimental study was done on 28 male Guinea pigs with 380±40g weight.for separating 3mm of saphenous vein rings and Measuring of rings nitric oxide released in preserving solutions: Krebs (K), Krebs plus propranolol (K+P), adrenaline (K+A) and verapamil (K+V) compaired with heparinized blood at 30, 45, 60 and 90 minutes after harvesting measured by micro plate Griess reaction. Rings also stained by H&E and examined by light microscopy to evaluate endothelial desqumation.
Results: Average concentration of nitric oxide (NO) in the Krebs plus Verapamil solution (K+V) Vs Heparinized Blood (HB), Krebs (K), Krebs plus Adrenaline (K+A) and Krebs plus Propanololol (K+P) revealed significant increase in NO release (P<0.05). The maximum NO measurement was 45 minuts after harvesting. Also histological study with H&E staining showed that endothelial layer was intact only in Krebs plus verapamil in compaired to control group, but in the other solutions the vascular intimal cells had suffered different degrees.
Conclusion: It seems that Krebs solutions containing verapamil has more efficiently to the proper functioning of the saphenous veins endothelium in animal modle.
