Abstract

Sialoprotein is one of the most abundant non-collageneous and phosphorylated glycoproteins in human. This protein plays an important role in the structure of human teeth. The aim of this project is to measure the amount of dentin Sialoprotein in the healthy and decay teeth to evaluate the variation in the teeth structure. In this investigation 50 decay teeth has been collected from patients referred to the clinic. The dentin was separated and placed in liquid Nitrogen. One gram of each dentin was washed with distilled water for 30 mins and subsequently the dentin was powdered, and relocated to the gaunidin-Hcl tris buffer, and incubated at 4°C for 48 hrs. The dentin powder was centrifuged at 3000 g for 20 mine. The supernatant was discarded, and the samples again was centrifuged at 10000 g. Finally one ml of this supernatant transferred to the sepharose column and washed with gaunidin-Hcl tris at 1 ml/min. The fractions obtained by chromatography was monitored by electrophoresis. The amount of decay teeth Sialoprotein was 17.23±1.45 ng/l and in the healthy teeth was 26.39±4.27 ng/l. The results from this study indicate that the Sialoprotein content in patient dentin decreased by about 1.5 time compared normal subjects.

Abstract

3 Beta-Hydroxy-delta-5-steroid dehydrogenase is an important key enzyme of steroid hormone biosynthesis, which is involved in catalyzing the conversion of pregnenolone to progesterone in the biosynthesis of steroid sex hormones. This is the fourth enzyme in the steroid hormone biosynthesis pathway to be identified as an autoantigenic target. Pregnenolone is incubated with several concentrations of whole testis homogenate. The reaction mixture containing the pregnenolone, NAD and iodonitrotetrazolium in 0.15 M Tris-HCL buffer (pH=7.7) and the enzyme extract from testis of rat was incubated for 1.5 h at 36°C. Absorbance at 495 nm was determined by a spectrophotometer. pH optimum and time incubation for this enzyme was 7.99 and 1.5 h respectively. The Kmax was 5.3×10^-6 and Vmax was 3.11×10^-6 nmol/mg/min. Our results were in good agreement with those reported previously.

Abstract

Background & Objective: Due to volatile property of monomer from Methylmethacrylate and its well known toxic side effects, in case of absorption via skin or respiratory system. Also, due to allergic reactions of it, consideration of suitable ventilation systems and using protective methods is very important. To decrease direct contact with it, the capabilities of technician and improves his health and work place, we investigate relationship between Methanol level and exposure to Methylmethacrylate. Materials & Methods: In a case-control (In vivo) study of 32 dental laboratory technicians in Babol and Sari before and after working and 32 normal subjects, urine samples were collected. Urine Methanol levels were analyzed by paper and gas chromatography. Results: Mean concentration of Methanol in case group (1.2 micro mol/l) was significantly more than control group (0.78 micro mol/l). Also, Methanol level was significantly higher after work (1.45 micro mol/l) when compared to before work (1.27 micro mol/l), (P<0.05). No relation between work history and urine Methanol level was observed. Conclusion: Due to significant increase of Methanol concentration during work in work place, the use of protective methods such as suitable ventilation, wearing mask and gloves, also, using vapor absorbing tubes can be useful in increasing dental laboratory workplace safety.

Abstract

Background & Objective: Glucose Amino Glycans (GAG) are unbranched polysacharides, major components of the basement membrane and play a key role in their molecular organization and function, also have an important role in the pathogenesis of some diseases such as hypertension. Hypertention is probably the most important health problem in several countries. But there is not yet a reliable indicator for early diagnosis of hypertension. The goal of this study was the measurement of serum and 24-h urinary GAG as an exact and early diagnostic marker. Materials & Methods: In this case – control study, 24-h urine and serum samples collected from the 53 patients and 38 persons as matched control normotensive group. Then amount of GAG was measured with spectrophotometery method. Results: Our findings showed that there is a direct relation between 24-h urinary GAG excretion and systolic blood pressure and it increases with increase of systolic blood pressure. Also amount of serum GAG increases in hypertensive patients in comparison with control group. Conclusion: This study showed that the concentration of GAG in sera and 24-h of urine samples increase in systolic hypertention.

Abstract

Background and Objective: Noninvasive methods have been proposed as surrogate markers for liver biopsy in recent years. It was shown that serum laminin level increases with the development for liver fibrosis The aim of this work was to determine serum laminin level cutoff point for predicting liver fibrosis, highlighting its diagnostic value and determining the effect of treatment on its level. Materials and Methods: In this case-control study during 2008-09, serum laminin levels in chronic hepatitis patients (n=62) and controls (n=20) before the beginning of the treatment and three times in a 2 month's interval i.e. 2.4 and 6 months after the beginning of the treatment- were compared by ELISA and stages of fibrosis were assessed according to the liver biopsy. Results: Mean serum laminin concentration in patients (91.9±20.9 ng/ml) was greater than the control (46.2±10.2 ng/ml, P<0.05). Serum levels of laminin in all stages of hepatic fibrosis were significantly higher than those of the healthy controls (P<0.05). A cutoff point of 52.0 ng/ml of laminin serum was obtained for the discrimination of patients with liver fibrosis than the healthy control showed a good sensitivity (96.8%) and specificity (80%). After 6 months of treatment, a gradual decrease in serum laminin level was observed, however the level was still higher than that of the healthy group (P<0.05). Conclusion: The findings of this study suggest that serum laminin level is a useful non-invasive marker of liver fibrosis due to strong positive correlation between serum laminin level and the degree of liver fibrosis.

Abstract

Background and Objective: Colorectal cancer is one of the most common cancers and considered to be leading causes of cancer death in the world. One of the problems with this type of cancer is its inability to be detected in early stage. The aim of present study was to evaluate the correlation between the serum levels of Fucosyltransferase-4 and galactin-3 in patients with early-stage of colorectal cancer.
Methods: This case-control study was done on 40 patients with early stages of colorectal cancer and 40 healthy subjects. ELISA method was used to measure the serum levels of Fucosyltransferase-4 and galactin-3. To examine the correlation between the variables, correlation test and Pearson correlation coefficient index were used.
Results: Fucosyltransferase-4 and galactin-3 levels were higher in patients with early-stage of colorectal cancer compared to healthy subjects. This difference was not significant. In patients with colorectal cancer, there was a significant relationship between the level of Fucosyltransferase-4 and galactin-3 (r=0.71, P=0.01).
Conclusion: Simultaneous measurement of Fucosyltransferase-4 and galactin-3 is useful in identifying early-stage of colorectal cancer as a non-invasive laboratory method.
