اثر تمرین تناوبی شدید و تداومی استقامتی بر بیان ژن‌های TRF2 و TERT بافت قلب موش‌های صحرایی نر پیر

زمینه و هدف: سن بزرگ‌ترین عامل خطر بیماری‌های قلبی - عروقی است که با کوتاه شدن طول تلومر همراه می‌شود و این در حالی است که فعالیت‌های ورزشی می‌توانند نقش مفیدی را در حفظ طول تلومر ایفا کنند. TRF2 و TERT به عنوان ژن‌های درگیر در مجموعه تلومریک در قلب نیز از پیری تاثیر می‌پذیرند و با کاهش بیان همراه می‌شوند که در نتیجه طول تلومر کاهش می‌یابد. کاهش طول تلومر نیز با افزایش مرگ سلول‌های قلب و بیماری‌های قلبی همراه می‌شود. این مطالعه به منظور تعیین اثر تمرین تناوبی شدید و تداومی استقامتی بر بیان ژن‌های TRF2 و TERT بافت قلب موش‌های صحرایی نر پیر انجام شد.

روش بررسی: در این مطالعه تجربی 24 سر موش صحرایی نر نژاد ویستار با سن 88 تا 96 هفته‌ای و میانگین وزنی 12±363گرم به مدت 6 هفته در گروه‌های کنترل، تمرین تداومی استقامتی (5 جلسه در هفته، شدت 60 تا 70 درصد بیشینه) و تمرین تناوبی شدید (5 جلسه در هفته، تکرار اینتروال با شدت بالا 80 درصد بیشینه در هفته اول و دوم و 90 درصد در هفته سوم و سپس 100 درصد سرعت بیشینه تا پایان تمرین) تقسیم شدند. بیان ژن‌های TRF2 و TERT به‌وسیله تکنیک Real time – PCR سنجش و پس از کمی سازی مقادیر بیان ژن با استفاده از فرمول پافل مورد بررسی قرار گرفت.

یافته‌ها: بیان ژن TRF2 گروه‌های تناوبی شدید و تداومی در مقایسه با گروه کنترل افزایش آماری معنی‌داری نشان داد (P<0.05). همچنین در بیان ژن TERT گروه‌های مورد مطالعه در مقایسه با گروه کنترل افزایش یافت و این افزایش از نظر آماری معنی‌دار نبود.

نتیجه‌گیری: به‌نظر می‌رسد شش هفته تمرین تناوبی شدید و تداومی استقامتی بتواند رشد و طول عمر سلول‌های قلبی را به‌وسیله حفظ طول تلوم از طریق افزایش ژن TRF2 تنظیم کند.
